    Value Added Grains for Local and Regional Food Systems

    Small grains provide numerous benefits for organic farming systems. However, because they have relatively low value and require special equipment, many organic farming systems do not integrate them. Heritage varieities of ommon wheat (Triticum aestivum ssp. Aestivum) and its antecedents spelt (Triticum aestivum, ssp. spelta), emmer (Triticum turgidum subsp. dicoccum) and einkorn (Triticum monococcum) are the subject of a project that looks at their viability. Preliminary results of variety trials show that organic farmers have several promising small grain options. Improved management and appropriate equipment are barriers to adoption that the project aims to overcome. Viability will depend on overcoming marketing as well as production obstacles

    Abstract Several useful alien gene transfers have been reported from related species into wheat (Triticum aestivum), but very few publications have dealt with the development of wheat/barley (Hordeum vulgare) introgression lines. An overview is given here of wheat 9 barley hybridization over the last forty years, including the development of wheat 9 barley hybrids, and of addition and translocation lines with various barley cultivars. A short summary is also given of the wheat 9 barley hybrids produced with other Hordeum species. The meiotic pairing behaviour of wheat 9 barley hybrids is presented, with special regard to the detection of wheat– barley homoeologous pairing using the molecular cytogenetic technique GISH. The effect of in vitro multiplication on the genome composition of intergeneric hybrids is discussed, and the production and characterization of the latest wheat/barley translocation lines are presented. An overview of the agronomical traits (b-glucan content, earliness, salt tolerance, sprouting resistance, etc.) of the newly developed introgression lines is given. The exploitation and possible use of wheat/barley introgression lines for the most up-to-date molecular genetic studies (transcriptome analysis, sequencing of flow-sorted chromosomes) are also discussed

    Drought Resistance And Productivity Of Wheat And Soybean Isogenic Lines With Different Photoperiodic Sensitivity

    The results of the study of drought tolerance of isogenic (NILs – near isogenic lines) by genes PPD (photoperiod) wheat lines and isogenic by genes ЕЕ (early maturation) soybean lines, that control the photoperiodic sensitivity are presented. In field experiments the photoperiodic sensitivity of the lines when grown under natural long days (16 hours at a latitude of Kharkov) and under artificial short-day (9 hours) is determined. The results showed that line PРD-D1A and PPD-A1a wheat and soybean lines L 71-920 had a weak photoperiodic sensitivity (weak PPDS) and line PPD-B1a wheat and soybean lines L 71-920 - strong photoperiodic sensitivity (strong PPDS). Wheat and soybean lines with weak PPDS were more productive. When simulating drought action on seed germination (20% strength mannitol solution - rapid method), it was showed that the seeds of soybean and wheat lines with weak PPDS have a higher germination than seeds of the lines with strong PPDS. When simulating soil drought (30% FMC – field moisture capacity of the soil) under growing experiment, it was revealed that the biomass accumulation indices of plants, leaf relative water content (RWC) and proline content in leaves lines with weak PPDS were higher than in the photoperiodic lines with strong PPDS. So, all used methods for determining drought tolerance showed that the low photoperiodic sensitivity lines are more resistant to drought.It is assumed that wheat genes PPD and soybean genes EE can participate in the formation of resistance to drought. Genotypes with low photoperiodic sensitivity should be used in breeding soybean and wheat drought resistance

    Zenon’s flour: grains of truth from Tel Kedesh

    According to one of the Zenon papyri, In 259 BCE the Ptolemaic courier Zenon stopped at the site of Kedesh, located today in northern Israel, to pick up some flour. In our excavations at this site from 1999-2011, we uncovered an enormous public administrative building with several storerooms filled with large jars. In one room fourteen locally made storage jars lined the walls. Phytoliths taken from the jars turned out to be identifiable as Triticum aestivum, commonly known as bread wheat. This may allow a scientific identification of “Syrian wheat,” a strain first mentioned in third century BCE Egyptian papyri as part of a package of agricultural innovations introduced by Ptolemy II Philadelphus.Accepted manuscrip

    Current taxonomic composition of European genebank material documented in EURISCO

    Taxonomy plays an essential role in genebank documentation. It is often the first level at which users search material, and it determines the protocols used in the management of collections. Especially, when plant genetic resources information is pooled in systems such as EURISCO, the European catalogue of ex situ plant genetic resources, problems regarding technical handling of taxonomic nomenclature, such as lack of standardization and low quality of data, become apparent. These problems were studied by analysing the content of EURISCO and mapping the taxon names in EURISCO on those used in the United States Department of Agriculture genebank system GRIN-Tax. Thus, the number of spelling errors and the level of standardization could be quantified and improved. An analysis of the content of EURISCO was made, showing a highly unbalanced distribution over crops: 50% of the accessions belong to ten genera only. Mapping EURISCO on the crops listed in Annex 1 of the International Treaty on Plant Genetic Resources for Food and Agriculture showed that 67% of the accessions in EURISCO belong to crops in that list

    Asymmetric somatic plant hybridization : status and applications

    To create asymmetric somatic hybrids, the genome of the so-called donor protoplast is fragmented prior to protoplast fusion. As a result, only a limited amount of the donor genome is transferred to the fusion product. This technique can circumvent some commonly observed problems related to symmetric fusion and offers a practical breeding tool for asexual hybridization. Genomes are typically fragmented by irradiation, microprotoplast production or application of metabolic inhibitors such as iodoacetamide. Irradiation and microprotoplast production fragment the nuclear genome, whereas iodoacetamide inactivates the cytoplasmic genome. It can therefore be used to introduce cytoplasmic male ste- rility, an important practical application. For hybrid verification and genome characterization, molecular markers and cytogenetic techniques are applied.     Detection of flour or farina from Triticum aestivum in macaroni by starch-gel electrophoresis of water-soluble proteins

    A water-soluble protein component, A, has been found which appears without exception at a higher concentration in Triticum aestivum than in T. durum. Quantitation of A is achieved by referring the height of densitogram peak A to the height of peak B, which corresponds to a second component that is present in all varieties studied from both species. Extreme valúes of peak height ratio (PHR) in both species are tentatively established. On the basis of these valúes, the máximum and minimum possible contents of T. aestivum in a mixture are expressed as a function of PHR. All the varieties of T. aestivum studied are detected when they contribute more than 60% to the mixture. In over 90% of all possible binary combinations between varieties of T. aestivum and T. durum, the máximum proportion of undetected T. aestivum has been 30%

    Unique and conserved MicroRNAs in wheat chromosome 5D revealed by next-generation sequencing

    MicroRNAs are a class of short, non-coding, single-stranded RNAs that act as post-transcriptional regulators in gene expression. miRNA analysis of Triticum aestivum chromosome 5D was performed on 454 GS FLX Titanium sequences of flow sorted chromosome 5D with a total of 3,208,630 good quality reads representing 1.34x and 1.61x coverage of the short (5DS) and long (5DL) arms of the chromosome respectively. In silico and structural analyses revealed a total of 55 miRNAs; 48 and 42 miRNAs were found to be present on 5DL and 5DS respectively, of which 35 were common to both chromosome arms, while 13 miRNAs were specific to 5DL and 7 miRNAs were specific to 5DS. In total, 14 of the predicted miRNAs were identified in wheat for the first time. Representation (the copy number of each miRNA) was also found to be higher in 5DL (1,949) compared to 5DS (1,191). Targets were predicted for each miRNA, while expression analysis gave evidence of expression for 6 out of 55 miRNAs. Occurrences of the same miRNAs were also found in Brachypodium distachyon and Oryza sativa genome sequences to identify syntenic miRNA coding sequences. Based on this analysis, two other miRNAs: miR1133 and miR167 were detected in B. distachyon syntenic region of wheat 5DS. Five of the predicted miRNA coding regions (miR6220, miR5070, miR169, miR5085, miR2118) were experimentally verified to be located to the 5D chromosome and three of them : miR2118, miR169 and miR5085, were shown to be 5D specific. Furthermore miR2118 was shown to be expressed in Chinese Spring adult leaves. miRNA genes identified in this study will expand our understanding of gene regulation in bread wheat

    Heterogeneity of wheat endosperm proteolipids (CM proteins)

    Proteins extracted with CHCl3-MeOH from wheat endosperm have been fractionated by Sephadex G-100 and the 15 000–20 000 MW range fraction, designated CM protein, has been examined by combined electrofocusing (pH range 5–8) and electrophoresis (pH 3.2) and the heterogeneity of the electrophoretic components has been ascertained. It has been shown by joint mapping and by sequential extraction that CM proteins are extracted by 70% EtOH but not by H2O, although they can be made water-soluble after dialysis against an acid buffer, pH 3.2, 3 M urea, without losing their solubility in CHCl3-MeOH mixtures. It is concluded that CM proteins fit the definition of a Folch—Lees proteolipid. The Triticum aestivum (genomes ABD) map can be reconstructed by mixing T. durum (AB) and Aegilops squarrosa (D).
